The purpose of this project is to optimize a method to allow a comprehensive identification of the proteomic profile of two organelles from wheat endosperm, endoplasmic reticulum and Golgi Complex. The method demonstrated a successful technique to enrich the target proteins and could serve as a model for investigating various complex organelles in crops. Improving crops quality requires an attention to the biochemical process of its development and knowledge of the machinery involved in this development. The protein synthesis machinery involves both organelles which determine storage proteins quality during endosperm development. The rate of reserve material accumulation varies during the grain filling stages.The constitutive proteins of the ER and GC are speculated to undergo changes in function, and a temporal change in protein expression during these stages assumed to be related to wheat protein quality. Consequently, the identification of the organelles proteins could contribute to the process of genetic modification intended to improve the crop quality.
