Presbycusis (age-related hearing loss) can result from various cochlear pathologies.The main aim of this master's thesis was to produce a suitable transplantable matrix which could be inserted into the lateral wall, by promoting the growth of fibrocyte cultures on a 3D matrix composed of collagen. This experiment was divided into two main sections which the first section included a comparison of fibrocyte cultures grown on a 3D matrix with 2D culturing non-coated wells or collagen coated coverslips in which cell growth and their morphology was taken into account. The second section of this experiment was the structural characterization of the collagen gel properties using the analySYS software.The results indicated that majority of fibrocyte cultures grown in wells on 2D surfaces coated with coverslips were typically spindle-shaped and flattened, whilst those grown on collagen I 3D gels tend to be small, more elongated with long processes, which form large clumps, or single-rounded individuals.For each labelled section from collagen-gel map, quantitative analysis was performed in which included the measurement of collagen-fibres diameter, pores-diameter and fibre density crossing.
