The present work aims to prepare and characterize mucoadhesive tablets of Atenolol using different buccoadhesive polymers such as chitosan, carbopol 934, Sodium alginate and HPMC K4M in combinations. The formulation of buccal tablet dose is released at a slow rate due to the presence of the polymers of different concentrations. All the buccal tablet formulations were subjected to pre-compression and post-compression evaluation. In-vitro drug release from the formulations was studied using buffer pH 6.8. From in vitro studies formulations F10, F16, F17, F18 were selected for mucoadhesive studies. Formulations F16, F17, F18 were selected for ex vivo permeation studies using buffer pH 7.4. After all studies formulation F18 containing chitosan, carbopol 934 and HPMC K4M in the ratio of 4: 3: 1 selected as optimized formulation showed zero-order release with optimum buccoadhesive strength and exhibited optimum drug release. FTIR results showed no evidence of interaction between the drug and polymers. The assay of the formulations revealed that the drug content was within the limits. The results indicated that delivery of Atenolol to the systemic circulation via the buccal route.
