Transfection in Vitro by using the Cancer Cell Line Model, we elected for our project three type of prostate cancer cell line from ATCC cell line company which are (PC3, DU145 & LNCaP ) cells, and use a reporter gene which is the ELK1 gene, transfection of gene plasmids was done by using the Liposome also from Lipofectamine 2000 liposome from Invitrogen company using tow type of the ELK1 gene 1st is the normal ELK1 gene ( control ELK1) 2nd is the Knock down ELK1 gene ( sh ELK1), for each type of test on cell lines we divide into two major group 1st is the (sh ELK1) & 2nd is (control ELK1) for the furthered comparison between these two group to show our reporter ELK1 gene effect on cell line cells activity. The 1st thing that was done is the Immuno Histo Chemistry IHC for prostate cancer cases for detect the presence of our Target gene ELK1 in the normal prostate cancer cells, that showed there is a presence of the ELK1 in prostate cancer case. Then we starter with working on the test that detect cell line activity in Vitro, the 1st was the WH assay (Wound Healing Assay) that show there is an very strong effect of the ELK1 gene on the wound healing process.
