Culturing of plant tissues and its ability to proliferate has enabled to increase the knowledge in many science areas including differentiation, tissue division, nutrition and transformation. In most instances, efforts have been directed towards providing aseptic conditions rather than getting into deep-dive science components associated with the type of contaminants, physiological status of inhabitant plant tissue, growing and growth conditions etc. The area of tissue culture has been progressed as an art rather than science which necessitates exploring the elements of scientific intricacies with much needed clarity for rendering solutions to complex problems. The present review highlights the importance of science component at different growth stages in the whole gamete of this most widely practiced plant technique which otherwise sparsely dealt in the past. Furthermore, an attempt has also been made to enlist the challenges linked in developing haploid & DH plants and possibleways to address them from research experiences of our own and from others.
