He graduated from Monroe County High School in 1990, and soon after, he pursued a degree in Agriculture from Western Kentucky University, graduating in 1993. During his time at university, Comer was actively involved in student leadership, notably serving as president of the Kentucky Future Farmers of America. After college, Comer, alongside his family, established James Comer Jr. Farms, a 2,300-acre farm, and he later co-founded Comer Land & Cattle Co. His work in agriculture didn't stop there; he also spent 12 years as a director for South Central Bank. From 1999 to 2000, he served as the president of the Monroe County Chamber of Commerce, marking his early entry into public service.
