Metarhizium guizhouense Strain CQMg135 is a promising entomopathogenic fungus. We genetically modified the M. guizhouense for further commercial developement a new myco insecticide as biocontrol agent to control white grub and other pest insects.But released entomopathogenic fungus how act in the soil and how long recombinant strains can survive under complex environment,it is not clearly known yet. So,for fruitful monitoring of the entomopathogenic fungus action during post inoculation period. We applied ATMT to transform green fluorescent protein(GFP) and enhanced green fluorescent protein(EGFP)genes into M. guizhouense(CQMg135). In two-chambered pot experimental, we also studied persistence and mobility of the genetically engineered strain. We created artificial barrier(porous divider wall covered with nylon mesh)in between tested strain and host plant and found that the tested strain(both recombinant and wild type)can overcome artificial barrier and successfully colonize in peanut root zone. After two months of inoculation, the propagules of the released strains were re-isolated from inner and outer rhizosphere soil titers which was almost similar with original inoculation.
