The present investigation entitled "Molecular Characterization of Parental Lines and Genetical Studies in Crosses of Cucumber (Cucumis sativus L.)" was carried out at Vegetable Research Centre and molecular characterization was performed in the NAIP Lab, Department of Vegetable Science Lab, and Cytogenetics and Plant Molecular Biology laboratory, Department of Genetics and Plant Breeding of the University. The major objectives of the study were to know variability parameters, heritability, genetic advance, determine of heterosis and combining ability in crosses between gynoecious and monoecious genotypes with the help of Line x Tester analysis and characterize parental genotypes using SDS-PAGE and ISSR marker. The study includes three gynoecious Pgyn-1, Pgyn-4, Pgyn-5 and ten monoecious genptypes i.e. PCUC-8, Pant Khira-1, US-832, PCUC-83, PCUC-15, PCUC-25, PCUC-35, PCUC-126, PCUC-208, Punjab Naveen. These parental lines were crossed to produce thirty F1s. Thirty F1s along with parents were evaluated in the next season to study heterosis and combining ability. The observations were recorded on five randomly selected plants for twelve yield and its contributing characters viz.
