Its southern terminus is at the Mason-Dixon line south of Wellersburg, and its northern terminus is at West High Street (formerly US 219) in Ebensburg. Along its route, PA 160 crosses the Eastern Continental Divide and passes over CSX Transportation's Sand Patch Tunnel.
