Use of microtuber is advantageous than microplants but dormancy problem of microtuber limits its production and use in the laboratory and the field respectively. This book provides a proven solution of dormancy breaking of freshly harvested microtuber with 5 to 7 days. A series of experiments in relation to production and breaking of dormancy of potato microtubers were conducted during 2000-2003. Nitrogen at 45 and 60 mM with 300-500 mg/l CCC or 30-50 mg/l coumarin or 5 mg/l BAP or 10% sucrose produced higher proportion of large size (200 mg) microtubers after 105 days. Treatment of intact microtuber with GA3 (0, 5, 10 and 15 mg/l) did not show any noticeable effect on dormancy breaking, while soaking of peeled microtubers treated with 0.1-1.0 mg/l GA3, 0.1 to 1.0 mg/l IAA and 0.01 to 0.1 mg/l Kin for 1 to 25 min and put on water soaked cotton pad containing in petriplates showed 90% sprouting within 7 days. Suberisation of peeled microtubers after treatment had no effect on sprouting but was safe from rot, while no rot of microtuber was observed while handling aseptically. This book would be useful for the tissue culture laboratory as well as basic seed potato growers.
