The prevalence of multi drug resistant (MDR) E. coli in food items (chicken meat) is of great public health importance. Retail chicken meat is major spreading factor of MDR E.coli infections in humans. This book focuses on MDR E. coli Plasmid DNA, its molecular profiling and curing. Molecular profiling comprises of extraction of plasmid through Phenol-Chloroform Isoamyalcohol method and determination of plasmid number and size in a particular strain by separation on Gel Electrophoresis (0.8%). Plasmid curing is done by exposing MDR strains to sub inhibitory concentration of chemical (SDS) and physical agents (heat). The confirmation of plasmid curing is done by replica plating and antibiotic susceptibility testing. The aim is to determine the relation of plasmid with antibiotic resistance pattern i.e. The antibiotic resistance is present either on plasmid (cured by plasmid curing) or chromosome.
