The SOS plasmid consisted of a vector,pOPRSVI/MCS, ligated to DNA oligonucleotidescorresponding to the SOS peptide of interest.Different SOS plasmid concentrations, ranging from1ug/uL to 5.138ug/uL, were either co-microinjectedwith Val 12 p21 or microinjected into oocytesfollowed by incubation in insulin or inprogesterone. At the 48th hour time point, theresults showed that the 3.225ug/uL SOS plasmid gavethe most inhibition on the oncogenic pathway whilethe wild-type p21 pathway was unaffected by thisconcentration. This data suggest that the 3.225ug/uL SOS plasmid is a specific inhibitor of theoncogenic ras-p21 pathway.
