De Paolis was born in Sonnino, and became a member of the Congregation of the Missionaries of St. Charles on 4 October 1958. He sent to Rome to complete his studies, he received his doctorate in canon law in the faculty of canon law at the Pontifical Gregorian University, a licentiate in theology at the faculty of theology at the Pontifical University of St. Thomas, and a law degree La Sapienza University in Rome. He was ordained to the priesthood on 18 March 1961, and taught canon law at the Pontifical Gregorian University in Rome.
