With this study, statistical analysis revealed that weight loss, grooming, wet dog shakes, head shakes, abdominal constriction, facial fasciculation, ptosis and piloerection. in rats occurred after termination of administration of 0.2 mg/kg and 0.4 ,mg/kg of benzhexol, pd, for 10 days. This study, also, reported that administration of 0.2 mg/kg of benzhexol, pd, for 10 days caused conditioning place preference in the rats. Also, rats consumed voluntarily benzhexol solution. Benzhexol caused elevation of the brain dopamine level, and termination of benzhexol administration caused reduction of brain dopamine, which is the cause of withdrawal symptoms. Administration of chlorpromazine and diphenhydramine (but not galantamine) re- increased the brain dopamine level in the rats. Administration of chlorpromazine and diphenhydramine (but not galantamine) after termination of benzhexol, reduced significantly benzhexol withdrawal symptoms in and consumption of benzhexol solution in rats. Benzhexol caused dose dependent neuronal degenerative changes in the neocortex and hippocampus areas in the rat brains. Benzhexol induced Straub tail and automutilation in rats.
