The MGMT gene produces DNA repair protein that is confirmed to confer resistance against cellular toxicity by alkylating agents. Therefore, intact MGMT protein in relation to TMZ therapy provides therapeutic detrimental protective impact and conversely is confirmed not to be present when MGMT protein is lost owing to epigenetic silencing of its promoter Methylation as seen in various tumors in particular glioma, thus discerning cells more sensitive to alkylating agents.In the present study, we investigated the molecular status of IDH2 and IDH2 gene in Glioma patients. We analyzed the mutational spectrum of the coding exons 4 of the IDH1 and IDH2 gene by direct sequencing. Wealso analyzed the promoter hypermethylation of MGMT gene by MS-PCR and its expression profile by Immunohistochemistry. Polymorphic analysis of, ACE, MTHFR and GSTP1 gene was also analyzed by PCR and RFLP.
