In the course of this work, we were interested in the development and technical validation of XM by CMF, which enabled us to deduce the following conclusions:Concerning the choice of TN, we found that NHP is the best control that can be used. The choice of negative sera in this pool was not based on the criteria of male sex and AB blood group. We plan to take these into account when building a new NHP. For PT, we compared the results provided by a positive serum and a pool of strongly positive sera.In the pure forms of the sera tested, a prozone effect was observed. This is due to the high concentration of anti-HLA Ac, which means that caution should be exercised when using this type of serum as a PT. However, the results were comparable for the poly-specific positive serum diluted at 1/10 and the pool of positive sera diluted at 1/2. We opted for the use of a pool of positive sera for its wider coverage of HLA antigenic specificities.
